Morphology: epithelioid cells growing adherently in monolayers
Medium: 80% mixture of DMEM and Keratinocyte SFM (at 1:1) + 20% h.i. FBS
Subculture: seed out at ca. 1-2*10⁶ cells/80 cm² flask; split confluent culture 1:5 once or twice a week using trypsin/EDTA (wash cells twice with PBS before adding trypsin/EDTA and apply trypsin/EDTA for up to 20 min. to detach cells); cells grow slowly during the first 1-2 weeks after thawing​​​​​​​
Incubation: at 37 °C with 5% CO₂
Doubling time: ca. 48 hours
Harvest: cell harvest of ca. 5*10⁶ cells/80 cm²
Storage: frozen with 70% medium, 20% FBS, 10% DMSO​​​​​​​​​​​​​​
